For 1 mg/L chloramine concentration, activation energy for diffusion of SBR and natural rubber were computed to be 51.13 kJ/mole and 77.29 kJ/mole, respectively. Functional metagenomics to discover antibiotic resistance genes in natural environments. $49.99 Despite the threat posed by antibiotic resistance in infectious bacteria, little is known about the diversity, distribution, and origins of resistance genes. Soil and insects may be important environmental reservoirs of antibiotic-resistant bacteria, but the significance of these reservoirs has not been explored. We undertook functional metagenomic analyses of a remote Alaskan soil and of culturable bacterial isolates from gypsy moth midguts to investigate antibiotic resistance genes in two environments with different potential to affect human health. The Alaska site is on an island in the Tanana River in the Bonanza Creek Long-Term Ecological Research Station near Fairbanks. We selected 12 gigabases of DNA in eight metagenomic libraries for genes encoding resistance to beta-lactam antibiotics. We report that this soil is a reservoir for diverse genes encoding beta-lactamases that function in Escherichia coli, including ancestral beta-lactamases and the first bifunctional beta-lactamase. In contrast to soil, which is largely sedentary, gypsy moths are an invasive species in the northeastern United States that feed on a wide variety of plants as they move westward. To identify antibiotic resistance genes associated with gypsy moth midgut bacteria, we built metagenomic libraries from a pool of 44 cultured isolates from wild and lab-reared gypsy moth larvae. We identified genes encoding three types of proteins among those responsible for resistance to diverse antibiotics: an efflux pump, a transcriptional regulator, and an extended-spectrum beta-lactamase. Our findings suggest that even in the absence of any obvious selective pressure imposed by anthropogenic activity, a soil microbial community harbors unique and ancient beta-lactam resistance determinants, and an insect midgut community carries genes that direct diverse mechanisms of antibiotic resistance. Stem cell-based composite cartilage tissue engineering using nanobiomaterials. $69 Degeneration of articular cartilage is one of the most chronic health problems, with poor ability to self-repair. Tissue engineering offers feasible strategies for regeneration of articular cartilage tissue, such as engineering biomaterial scaffolds seeded with human mesenchymal stem cell (hMSC)-derived chondrocytes. It is crucial to mimic the distinct physiological zonal organization of cells and extracellular matrix (ECM) in engineered cartilage constructs to accurately restore structure and function. Furthermore, engineered tissues with cellular microenvironments and nanoscale architectures analogous to natural tissue can regulate cell differentiation, adhesion, and orientation. To engineer thin superficial zone where cells and ECM are oriented parallel to articular surface, oriented electrospun polycaprolactone scaffolds comprised of nanofibers (∼500 nm diameter) were used to guide hMSC orientation and adhesion. Viability and orientation of hMSCs were maintained for 35 days. Fluorescence visualization of microfilaments showed cytoskeletal reorganization of elongated hMSCs on nanofibers, and actin-containing tether-like structures appear to mediate cell-nanofiber adhesion. Chondrogenic differentiation was induced and cartilage-specific markers were analyzed, indicating that hMSC-derived chondrocytes on oriented PCL nanofibrous scaffolds may be ideal to mimic superficial zone. To engineer transitional zone where spherical cells and ECM are randomly distributed, hMSC-seeded 3D collagen type II-based hydrogels mechanically reinforced by multi-walled carbon nanotubes (MWCNT, 200-500 nm diameter) were used. MWCNT-collagen hydrogels demonstrated significant increase in mechanical strength assessed by atomic force microscopy, and thus offer more biocompatible alternative to chemical crosslinking.
